This was the first-in-human Na-ASP-2 hookworm-vaccine study in adults without previous hookworm infection. The randomized, double-blind, placebo-controlled trial enrolled 36 healthy adults. Three dose cohorts each contained nine vaccine recipients and three saline recipients; injections were scheduled on days 0, 56, and 112. 123
The original report found antibody responses after vaccination and no serious adverse events in this study. Injection-site reactions were generally mild or moderate, and delayed local reactions were reported. These are safety and immunogenicity findings, not proof that the vaccine prevented hookworm infection or disease. 2
Population matters: results in previously unexposed adults must not be merged with a different trial in previously infected participants. A vaccine's ability to generate antibodies and its suitability for an exposed population are distinct questions. 2
